Use of sorological and molecular techniques in the distinction of vaccinated and naturally infected dogs by Leishmania (l.) infantum chagasi

Abstract

Serological methods are used in canine surveys and the methods currently employed do not distinguish infected dogs from vaccinated ones. One of the foundations used to control canine visceral leishmaniasis (CVL) is euthanasia, but there are other interventional methods such as the use of collars impregnated with pyrethroids and anti-Leishmania vaccines that could be used in LV control programs. One of the problems in using the vaccine in public health is that during canine surveys it may be difficult to distinguish vaccinated dogs from those infected. From samples obtained from a previous work, supported by FAPESP, carried out in Panorama-SP city, we will evaluate if it is possible to distinguish vaccinated dogs from those naturally infected. Samples from six different intervention groups, collected at 0, 180 and 360 days, will be used. Samples of serum and DNA obtained from lymph nodes will be used. Serum samples will be evaluated by the following methods: Immunochromatographic test (DPP); ELISA L. major-like and ESA ELISA. Lymph node samples will be used to detect Leishmania infantum chagasi DNA using real-time PCR (qPCR) targeting the DNA of the kinetoplast mini-circle. As for the data analysis, the serological values of each test with its antigens will be compared to accuse those who demonstrate positivity in the reactions. DPP by table indicating the positive and negative results and the ELISAs by means of the optical density. Real-time PCR will be performed in four analyzes: Amplification and dissociation curve, Spectrum and Component. It is hoped that the research will contribute to distinguishing vaccinated dogs from those infected. (AU)
