The anti-inflammatory effect of curcumin on renal ischemia / reperfusion injury in diabetic rats

Abstract

The presence of Diabetes Mellitus (DM) increases the susceptibility to acute renal injury caused by ischemia and reperfusion (I / R), which in clinical practice may coexist in diabetic patients under critical conditions. I/R and DM share common physiopathological mechanisms, involving dynamic physiopathology comprehending inflammation and oxidation. Curcumin, present in Curcuma longa L., is a natural anti-inflammatory that can attenuate aggravated I/R complications in DM. The objective of this study is to evaluate the effect of Curcumin on renal, hemodynamic and oxidative profile of diabetic rats submitted to I / R. Wistar rats, male and adult, weighing between 250-290 g, were randomized into four groups: Citrate; DM; DM + I / R; DM + I / R + Curcumin. All groups will be followed for 28 days. Physiological parameters such as feed and water intake, glycemia and kidney weight / animal weight ratio will be evaluated. Renal function will be assessed by inulin clearance, proteinuria, urinary flow, plasma and urinary creatinine. Renal hemodynamics will be assessed by measuring renal blood flow and calculating renal vascular resistance and oxidative profile by measuring urinary peroxides, urinary nitric oxide, thiobarbituric acid reactive substances, and thiols in renal tissue.
