Hepatotoxic, nefrotoxic and toxicogenetics analysis of animals implanted with polyamide membrane 12 incorporated with hydroxypatite

Abstract

In small animals, bone defects are commonly diagnosed and, for this reason, the search for new materials that aid in bone tissue repair (osteoinduction and osteoconduction) and total recovery of motor function is constant and justifiable in veterinary orthopedics. Thus, the objective of the present study is to evaluate the angiogenic and osteoinductive activity of the hydroxyapatite incorporated in prototyped polyamide 12 membranes (implant), when allocated to different intracorporeal sites and to investigate possible deleterious effects of this implant in non-target organs of the research. In the bioassays, 40 rats will be used, of which 30 will be submitted to anesthesia and surgery for allocation of three intracorporeal implants in each, at different sites (subcutaneous, omentum and bone) and euthanasia will be performed in five of these per period (7, 15, 21, 30, 45, and 60 postoperative days). In addition, five animals will comprise the negative control (GN) and five, the positive control (GMMS) for the toxigenicity tests, which will not be submitted to the surgical allocation of the implants. After euthanasia, blood (except GMMS) will be collected for the determination of urea, creatinine, hepatic enzymes and bone marrow in order to evaluate, respectively, the possible nephrotoxic, hepatotoxic and toxicogenic effects of intracorporeal implant. Afterwards, the intracorporeal sites implanted (except the GN and GMMS) will be collected for histological analysis and by electron microscopy. The results will be analyzed statistically by analysis of variance (ANOVA One-Way); in cases where Pd0.05, the means of treatment will be compared by the Tukey method, with the calculation of the minimum significant difference for ± = 0.05, with the use of the program Graphpad Prism.
