Physicochemical properties analysis of AH plus associated with antimicrobial substances

Abstract

The AH Plus is an endodontic cement considered a Golden pattern, which will execute the union between the interfaces gutta-percha and dentin walls, however, its action against microorganism is limited. The ambroxol (ABX) is a metabolite VII of bromhexine and has a known monoclinic and expectorant pharmacological action. The N-Acetylcysteine is a potent antioxidant and derivative of cysteine, it's a non-antibiotic compound, but it has antimicrobial action. Therefore, this study's objective is to evaluate AH Plus associate with 5% of N-Acetylcysteine and AH Plus associate with 5% of Ambroxol, about its physical properties (radiopacity, setting time, flow, solubility, compressive strength) and antimicrobial action. To evaluate radiopacity, we will use metal rings where experimental cement will be put and maintained. Those will be radiographed and processor manually, for subsequently digitized and analyzed in the program Digora 1.51. Setting time: Experimental cement poured in metal rings will be subjected to marking with vertical pressure, with Gilmore needles. Flow: The cement will be spatulate at the center of a glass plate where another one will be put on it, both with 100g, and after 10 minutes of the beginning of the spatulation, the weight will be removed, then, the longest and smallest diameter of the cementer will be measured with help of a digital caliper rule, and the flow will be an average oh both diameter. Solubility: 30 acrylic teeth will be filled with experimental cement. The analysis will be done with a comparison of the volume in mm³ by imagens of computerized microtomography (micro-CT). Antimicrobial action: on the metal rings filled with experimental cement and group control will be induced in vitro biofilm of Enterococcus faecalis. After a period of 21 days, the samples will be stained Live/Dead Technique-In Vitro Gen, and in sequence will be evaluated in a Confocal Laser Scanning Microscope. On the cement blocks, adhesion will be made of Ent. faecalis. The results will be submitted to D'Agostino and Pearson's tests fo verification of normal distribution.
