Evaluation of the effectiveness of treatments with phosphopeptide of casein-amorphous calcium phosphate and sodium trimetaphosphate on the remineralization of caries lesions: an in vitro study

Abstract

The aim of the present study was to evaluate in vitro the effect of different antiarrhythmic agents containing amorphous phosphopeptide calcium phosphate (CPP-ACP), sodium trimetaphosphate (TMP) and fluoride (F) and different treatment agents in remineralising caries lesions. Bovine enamel blocks (4 mm × 4 mm, n = 60) will have their enamel surface polished to determine the initial surface hardness (HS). Afterwards they will be submitted to the induction of artificial caries lesions and the post-demineralization hardness (SH1) will be determined. The experimental design was randomized and the blocks divided into 5 groups of treatments (n = 12): 1) Dentifrice without F / TMP / CPP-ACP (Placebo); 2) Dentifrice with 1100 ppm F (DF); 3) Dentifrice with 1100 ppm F + Neutral gel with 9000 ppm F (DF + Gel F); 4) Dentifrice with 1100 ppm F + Neutral gel with 4500 ppm F + 5% TMP (DF + Gel TMP) and 5) Dentifrice with 1100 ppm F + MI Paste Plus®. For groups 3 and 4 the gel will be applied once for 1 minute, initially to the study. Already for group 5 after treatment with DF, it should be applied to MI Paste Plus® 2x/day for 3 minutes. The blocks will be submitted to 6 pH cycling for 6 days. After the pH cycling, the post-cycling surface hardness (SH2) will be determined for the calculation of the percentage of surface hardness recovery (% SHR) and longitudinal hardness, to calculate the integrated loss of subsurface hardness (”KHN). The data will be submitted to the normality and homogeneity tests to use the statistical test most appropriate to the data distribution. Sigmaplot© software for Windows version 12.0 will be used with statistical significance at the 5% level.
