Abstract

The Fungal Bioluminescence Laboratory (FBL) of the Chemistry Institute/University of São Paulo (IQ-USP), through the Thematic project of FAPESP "Electronic Chemiexcitation in Biological Systems: Bioluminescence and 'Photochemistry in the Dark'", offers two Post-doctoral positions for candidates with a PhD degree obtained for a maximum of 7 (seven) years. It has been proposed to investigate mechanisms of radical or enzymatic generation of electron-chemiexcited species in biological systems, involved in damage of proteins and DNA thereby triggering pathogenic responses. Among them, we aim to study (i) the mutagenic cyclobutane dimerization of DNA pyrimidines in the absence of light and their quenching by sorbates, (ii) modification of hemeproteins (HRP, myoglobin, hemoglobin) in the presence of H2O2 with loss of peroxidase activity, (iv) search of new sources of singlet oxygen, specifically acrolein present in thermoprocessed food and formylhydrazine found in some poisoning mushrooms (e.g., Gyromitra esculenta), and (iv) enzymatic generation of triplet carbonyl compounds in model systems for oxidative and carbonyl stress. Kinetic studies, isotopic labelling and conventional spectroscopic, photochemical and photophysical methods are experimental approaches in the development of the project. Our project aims to accumulate knowledge underlying the chemiexcitation field in biomedicine, develop bioanalytical tools, and foster international collaborations.
