Histomorphometric analysis of the effects of the administration of the probiotic Bifidobacterium animalis subsp. lactis HN019 on experimental periodontitis progression in rats submitted to chemotherapy

Abstract

Chemotherapy treatment increases the severity of periodontitis and compromises treatment outcomes. Probiotics have emerged as an alternative for the treatment of oral pathologies, modulating the host immune response and modifying the bacterial environment. The aim of the present study is to evaluate the effects of Bifidobacterium animalis subsp. lactis HN019 probiotic administration on experimental periodontitis (EP) progression in rats submitted to chemotherapy (5-fluorouracil - 5-FU) through histomorphometric analysis. 80 male rats will be used, divided into the following groups: control (C); treatment only with 5-FU (5FU) (60 and 40 mg/kg); treatment only with PROB (HN019) (daily for 44 days, beginning 30 before EP induction); treatment with 5-FU and PROB (5FU-HN019); only EP (EP) (ligature on inferior first molars, left for 14 days); EP and treatment with 5-FU (PE-5FU); EP and treatment with PROB (PE-HN019) and EP and treatment with 5-FU and PROB (PE-5FU-HN019). All animals will be euthanized 44 days after the beginning of the experiment and hemi-mandibules will be collected for the measurement of the alveolar bone level and the attachment loss in the distal site of the first molar, and the area with no bone in the furcation. For normal distribution data, Variance Analysis (ANOVA) followed by the post-hoc test Tukey will be used. For data with non-normal distribution, Kruskal-Wallis followed by the posthoc test of Dunn will be used. It is expected with this study to demonstrate that the use of probiotics could be considered as an important tool in periodontal disease prevention in individuals under chemotherapy.
