Extraction and characterization of ULVAN: platform for preparation of biomimetic systems particulates for intestinal permeation of hyaluronic acid and chondroitin sulfate of vegetable origin (Ulva lactuca)

Abstract

As seaweed of the order Ulvales are very common in the coastal region of all the oceans of the Earth, as well as in largest reservoirs of freshwater. The main kinds of this order of green algae are the kinds Ulva and Enteromorpha, which have the characteristic of rapid proliferation when they are found in eutrophic waters in coastal regions and in reservoirs. In this proliferation, the formation of a species of "green bloom" occurs, which causes the death of organisms present in these waters, because of hypoxia, that is, due to the decrease of oxygen levels in the water. In this way, to perform the exploration of the algae biomass of these kinds, can solve the environmental problems caused by them. Ulva lactuca is one of the green algae that are present in the Brazilian coast in abundance, mainly in Santa Catarina's coast. This alga has many advantages in its exploration at research and development of pharmaceutical products. The main component of this seaweed is the Ulvan, one class of sulfated heteropolysaccharides that are withdrawn from the cell wall rich in hyaluronic acid and condroitin sulphate. Both are polymer of glycosaminoglycan that is composed of monomers of glucuronic acid and acetylglicosamine, join its are responsible for maintaining the homeostasis and the mechanical integrity of tissues and fluids that have structural functions and lubricants. OBJECTIVE: The objective of this project is to evaluate the viability of the use of seaweed Ulva lactuca as source of hyaluronic acid and chondroitin sulfate. Methods: This evaluation will occur through of extraction and of characterization the polysaccharides from this algae, of preparation and of characterization of the micro and nanoparticulates systems obtained with these polysaccharides and of evaluation the transmural permeation in the ex vivo model with the use the inverted intestinal sac method. Expected results: Thus, it is expected to confirm that the hyaluronic acid can be obtained from seaweed and that it has a greater bioavailability than those coming from the animals or from the biosynthesis process, that is, that its percentage of use by the organism is greater. For this confirmation, it is expected to obtain a nanoparticulate system with the globular format and composed by the mixture of chondroitin calcium sulfate and calcium hyaluronate. (AU)
