Abstract

The present project will use the combination of different materials to advocate the construction of scaffold using polycaprolactone (PCL) and polyethylene glycol (PEG) combined with a deoxyribonucleic acid (DNA) hydrogel applied as a human fetal osteoblast cellular transport system (hFOB 1.19) ATCC (CRL-11372), used in bone regeneration. In addition, a second strategy of direct application under the scaffolds of mesenchymal stem cell lines derived from human bone marrow (HBMS) ATCC (PCS-500-012) will be used to obtain functional osteoblasts. aroused great interest due to their potential involvement in the tissue repair process. DNA-based hydrogels have the function of improving the cellular interaction with pre-fabricated scaffolds, homogeneously distributing the cells and thus mimicking the complex structure of the tissues in addition to improving the conditions of cellular interaction, limited in PCL-PEG scaffolds . In a previous work, it was demonstrated the potential of DNA application as high biocompatibility material, allowing the application to regenerative medicine. Thus, in the present project, we aim to: (i) optimize, evaluate the interaction conditions of hydrogel-cells and the molecular mechanisms of interaction and adhesion between hFOB cell line 1.19 and scaffold; (ii) to differentiate the lineage of mesenchymal stem cells (HBMS) leading to the formation of osteoblasts under the scaffold and preceding the development of scaffolds with effective clinical and experimental application and the possibility of commercial use of the present product.
