Effect of different concentrations of glycerol in frozen dog semen

Abstract

To ensure the feasibility seminal after cryopreservation, the presence of the means diluidores functioning as caps and suppliers of nutrients is extremely important. The glycerol is the crioprotetor most used for change the properties colligative water, decreasing the freezing point and increasing the survival of the cells sperm. Several works were performed with different concentrations of glycerol in frozen of semen dogs with distinct results, so it is necessary to further studies in the area. In this sense, the objective of this study will evaluate different concentrations of glycerol 5, 6 and 7% on the frozen of dog semen. Will be used 10 dogs adults between two to five years of different races. The ejaculates will be harvested by digital manipulation and after harvest the ejaculate will be divided into three rates that will be centrifuged and resuspendidas in thinner to freezing the basis of egg yolk (botudog®) with three different concentrations of glycerol, 5, 6 and 7%. Will be evaluated the characteristics of kinetic sperm through the system CASA and integridades membranes plasma and acrossomal, as well as the translocation of phospholipids membrane and evaluation of training sperm by flow cytometry in moments post thawing and after 60 minutes incubation the 370c. Data will be assessed statistically regarding the distribution of normality and then analyzed by tests parametric or non-parametric.
