Biological and physicochemical properties evaluation of a bioceramic-based intracanal medication BIO-C temp

Abstract

The aim of this study will be to evaluate the systemic response, biocompatibility, and bioactive potential and physicochemical properties of a bioceramic-based intracanal medication, BIO-C TEMP in comparison with the Calen paste. Polyethylene tubes will be filled with BIO-C TEMP or Calen paste and will be implanted in the dorsal subcutaneous of sixty male adult rats distributed into 3 groups: BIO-C TEMP (BIOG), Calen (CALG) and control (CG; empty tubes). After 7, 15, 30 and 60 days of implantation, the rats will be anaesthetized, and the blood will be collected by cardiac puncture, and the serum calcium, phosphorus and alkaline phosphatase levels will be measured by chemiluminescence method. Subsequently, the polyethylene tubes surrounded by tissues will be removed and fixed in formaldehyde. Longitudinal sections will be stained with Hematoxylin and Eosin (HE) for morphological analysis. The number of inflammatory cells in the capsules and the capsule thickness will also be obtained. Sections will also be subjected to immunohistochemistry for detection of interleukin-6 (IL-6), Vascular Endothelial Growth Factor (VEGF), Osteopontin (OPN) and osteocalcin (OCN) in the capsules. The von Kossa method will be performed for the detection of calcium deposits. For pH evaluation, polyethylene tubes (n = 10/group) filled with BIO-C TEMP and Calen paste will be immersed in the distilled water. After 12 hours, 1, 3, 7, 14, 21 e 28 days the pH of medium will be measured. Radiopacity of the materials will calculated in relation to the optical density of the aluminum. In order to evaluate the solubility, the acrylic teeth (n = 10/group) will be digitized with microtomography immediately after the root canal filling with BIO-C TEMP or Calen paste, and after 7, 15 and 30 days. The volumetric loss will be calculated by the difference between the initial and final volume. Forty human unirradicular teeth will be used to evaluate the possible presence of material residues after removal of endodontic medications. After root canal instrumentation, the root canal will be filled with BIOG or CALG (n=20/group). After 30 days, the medications will be removed by conventional manual irrigation or passive ultrasonic irrigation (n=10/group), the tooth will be longitudinally sectioned and with the help of a stereomicroscope will be quantified the BIO-C TEMP and Calen remaining adhered to the root dentine walls using the ImageJ Software. The quantitative data obtained will be subjected to appropriate statistical tests for analysis, with significance level considered of 5%. (AU)
