Abstract

Bioresorbable polymers with embedded ceramic nanoparticles can be used as biomaterials because of their physico-chemical characteristics, structural versatility, bioactivity and superior mechanical properties and have attracted significant attention to tissue engineering. Among the ceramic nanoparticles is the bioglass, which presents bioactive and mechanical properties, and ease in the incorporation of functional groups that cause physiological responses to bone cells and tissue regeneration in vivo. Several techniques can be used for the production of polymers aiming at the nano/micro fiber fabrication seeking the biomimetism with the extracellular matrix, we highlight electro-spinning. In this project the influence of the incorporation of the bioglass in polymeric scaffolds of PCL will be evaluated, aiming at bone regeneration in critical defects of tibias of rats. For the morphological characterization of the materials will be used: scanning electron microscopy (SEM), wettability analysis and x-ray diffraction. For the in vivo assays, critical bone defects (3mm) will be made in tibias of Wistar male rats, which will be filled with different biomaterials: a) clot (control group); b) PCL; c) PCL with micrometric particulate bioglass; d) PCL with nanoparticles bioglass. Both critical bone defectswill be will be filled with same material. After 04 weeks of surgery, the animals will be euthanized and the right tibiafragments will be assessed through radiographic, histologic and histomorphometic analysis, to verify the better way of developing bone regeneration due the influence of the biomaterial, meanwhile the left tibia will be submitted to flexion resistance test. The data will be statistically analyzed, initially by means of the normality test aiming the application of the appropriate statistical test. The level of significance adopted will be the conventional value of 5%.
