There are ~1000 Odorant Receptor (OR) genes in the mouse genome. Each olfactory sensory neuron expresses one allele from one single OR gene. The molecular mechanisms underlying OR gene expression is not understood. Our previous results have shown that olfactory neurons have a characteristic nuclear organization which differs from other cell types. Odorant receptor genes that are located on different chromosomes are frequently associated to large common constitutive heterochromatin block which are located in a central region of the nucleus. Interestingly, facultative heterochromatin is predominantly localized around the large constitutive heterochromatin blocks. In this project we aim to study the role played by facultative heterochromatin in gene expression in the olfactory sensory neurons. We also plan to determine whether non-coding RNAs play a role in this regulation. Transgenic P2-IRES-tauGFP mice, which express GFP in the olfactory sensory neurons that express the P2 OR gene, will be used in these experiments, together with in situ hybridization, RNA seq and bioinformatic techniques.
