Abstract

Protein corona is a phenomenon characterized by the spontaneous and nonspecific adsorption of proteins on nanoparticles' surface. This protein shell influences on the biological function of nanoparticles applied in Nanomedicine. Within this context, zwitterionic groups are among the compounds used to supress the protein corona formation. Nevertheless, in order to apply these nanoparticles to therapies and diagnostics, it is necessary to get a deeply knowledge about the interaction and internalization of them into cells. Considering these facts, this project's motivation is to study the interaction and internalization of silica nanoparticles (SiO2NPs) by human cells. For this, nanoparticles that induce the formation of protein corona and nanoparticles functionalized with zwitterionic compounds (absence of protein corona) will be analyzed. Among the studies to be performed in this project, the highlight is a detailed thickness and morphology protein corona characterization carry out by cryo-electron transmission microscopy. Moreover, transmission microscopy and X-ray tomography will allow to get a complete and fundamental view of the internalization process and localization of nanoparticles in human cells either in 2-D and in 3-D, respectively.
