Abstract

Mast cells (MCs) play a key role in the inflammatory response, tissue repair and defense of the body, and when activated they release their rich granules with inflammatory mediators, such as interleukins and growth factors, which may play an important role in bone tissue. Bone is a metabolically active tissue, subject to continuous remodeling throughout life, ensuring the maintenance of mechanical strength and systemic physiological homeostasis. Osteoblasts (OB) represent a small percentage of resident bone cells, and are known for their constructive function. They are responsible for the synthesis and secretion of the organic matrix, its maturation and mineralization. Although recent studies have addressed the role of MCs in osteogenesis, the exact interaction and mechanisms involved in this process is not yet fully elucidated. Therefore, the purpose of this study is to investigate the role of MCs on osteoblast activity from calvaria cells of spontaneously hypertensive rats, which present a deficit in the quality and density of bone tissues. For this, the culture of the culture supernatant mast cells sensitized and stimulated with IgE + DNP will be performed for possible evaluation of the role of the inflammatory mediators released by the MCs that influence the mineralization of the extracellular bone matrix. The prediction is to obtain in vitro results that may contribute to a better understanding of the role of MCs in bone remodeling associated with an epidemiologically relevant systemic disorder such as hypertension.
