Obesity is a multifactorial condition, considered one of the most serious problems of public health worldwide and is involved in an increased risk of developing chronic diseases, especially cardiovascular diseases. Individuals with obesity usually present evidence of cardiac remodeling, defined as the set of molecular, cellular and interstitial cardiac variations, in response to a given aggression. Matrix metalloproteinases (MMPs) and metalloproteinase tissue inhibitors (TIMPs) contribute to tissue remodeling in various physiological and pathological states, and the balance between them is essential for tissue remodeling of the extracellular matrix. In this context, the MMP-2 gene is highlighted, since it is significantly involved in cardiovascular remodeling, and its inhibitor TIMP-2. Furthermore, some exogenous factors, such as significant weight loss, can modify the expression of these genes. In this context, one of the treatments indicated for the cases of severe obesity is bariatric surgery, being Roux-en-Y gastric bypass the most used, considered a fundamental strategy for weight reduction and, thus, improvement in cardiac remodeling. Objective: To evaluate the expression of MMP-2 and TIMP-2 genes in women in the pre and postoperative period of bariatric surgery and the association of these genes with body composition and anthropometry parameters. Methods: Patients were selected and divided into two groups: Group 1, containing 30 women with grade III obesity in the preoperative period of bariatric surgery (G1A) and the same 30 women six months after the surgical procedure ( G1B). Group 2, containing 10 eutrophic women, as control group. Peripheral blood collection for RNA extraction, anthropometric data and body composition have already been performed. The biological samples will be submitted to an RT-qPCR, to evaluate the expression levels of the MMP2 and TIMP2 genes. For statistical analysis will be used the Shapiro-Wilk tests, ANOVA with post hoc of Tukey or Kruskal-Wallis and t test. The significance level for p <0.05 will be accepted, and all analyzes will be performed in the Statistical Package for Social Science software (SPSS version 17.0 [Inc. Chicago, IL]).
