Abstract

Cellular differentiation relies on differential transcription of genes. However, many fundamental questions about transcription control remains unsolved, for instance, which properties establish the interaction between transcription factors and their respective binding sites in vivo? Or, how does the interaction between transcription activator and repressor factors in gene regulatory regions happens? These questions our group investigate in the laboratory using the segmentation cascade which acts at the beginning of Drosophila melanogaster embryonic development as a model of studies. In this project we propose to investigate the activity of the transcription factor Tailless (Tll). Data from the laboratory show multiples targets of Tll including pair-rule and gap genes, not detected before. We will explore the fact biochemical data from genomic studies are available for Tll and we will cross these data with genetical data from laboratory. Also the distincts Tll PWM will be analysed and the most significative data will be compiled into a new PWM. Our goal is to investigate if there is a direct interaction between Tll and the regulatory regions of putative targets as well as predict possible binding sites with maximum accuracy. This study will provide a pioneer protocol that we intend to establish to investigate others segmentation genes, therefore, contribute to the understanding of Drosophila anterior region specification, precursor of the future head, and to generate models/hypothesis operating at the local regulatory net.
