Abstract

Incorporation into cell membranes is key for the action of photosensitizers in photomedicine treatments, with hydroperoxidation as the prominent pathway of lipid oxidation. Herein we shall fabricate Langmuir monolayers of dioleoylphosphatidylglycerol (DOPG), 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) and1,2-dipalmitoyl-sn-glycero-3-phospholcholine (DPPC) as cellular membrane models to investigate the adsorption of the photosensitizer hypericin and the photo-induced effects of lipid oxidation. The Langmuir monolayers of phospholipids containing different concentrations of hypericin will be made on ultrapure water subphase, characterized by surface pressure (À) vs. mean molecular area (A) isotherms, ellipsometry imaging, Brewster angle microscopy (BAM) and surface rheology. The lipid membrane loaded with hypericin will be further photo-activated to analyze the effects of lipid oxidation on the membrane properties, which may include the increase of the mean molecular area of the lipids, the cleavage of the carbonic tails and changes in the membrane viscosity. This BEPE proposal is part of the scientific initiation project (2017/13404-3) entitled "Lipid photo-oxidation mediated by the xanthene eosin Y into cellular membrane mimetic system based on Langmuir films", which has been carried out at the FCL-UNESP, Assis, SP.
