Investigation of reproductive parameters of male rats exposed to anti-inflammatory ibuprofen in pre-puberty

Abstract

Ibuprofen is a nonsteroidal anti-inflammatory (NSAID) non-selective inhibitor of the enzyme cyclooxygenase (COX-1 and COX-2), which reduces the production of prostaglandins (PGs). As recent studies have reported a lot of PG functions in the male gonad, COX inhibiting drugs may negatively impact the male reproductive system. In addition, analgesics of the NSAIDs class may be considered as endocrine disrupters with antiandrogenic effects, being capable of change the reproductive function of both men and animals from fetal to adult life in both sexes. Because ibuprofen is one of the most prescribed anti-inflammatory drugs in pediatric practice (critical period of reproduction), it is necessary to investigate the reproductive effects of ibuprofen after exposure during childhood and adolescence. This project aims to evaluate the effects of exposure to ibuprofen, in juvenile to peripuberty phases on reproductive parameters of male rats in adulthood. For this, we will use Wistar male rats (n = 40; 23 days old) distributed in 4 groups: Control (CMC - carboxymethylcellulose 1%); and three groups that will receive three different doses of ibuprofen: 2.4; 7.2 or 14.3 mg/kg/day. The treatment will occur for 31 consecutive days, orally (gavage). Clinical signs of toxicity and the puberty set up of these animals will be evaluated during treatment. At 85 days of age (adulthood), fertility test, sexual behavior, hormonal dosage and sperm parameters will be evaluated. (AU)
