Abstract

As a way to lessen dependence on fossil fuels, second-generation ethanol, obtained through the enzymatic hydrolysis of plant biomass, has been the focus of several scientific investigations. However, the recalcitrance of the plant cell wall and the high price of the hydrolytic enzymes mean that its complete and efficient degradation is still a challenge to be overcome for its economic viability. Therefore, the main goal of this project is to evaluate the influence of the deletion of transcription factors in Aspergillus nidulans, on the production/secretion levels of plant biomass-degrading enzymes. The transcription factors to be deleted are related to cellular stress response to heterologous protein secretion, which has been carefully selected from RNA-seq data. This study aims to find a mutant strain with increased production of plant biomass-degrading enzymes. Additionally, it aims to generate data for a global view of the cellular reprogramming process in A. nidulans. In conclusion, these goals may provide researchers with resources to better propose rational engineering strategies that improve the secretion process and thus, reduce the cost of producing enzymes.
