The images obtained by fluorescence microscopy are widely used in many researche areas, including with application well-established in clinical areas (diagnostic). On the other hand, the surface-enhanced Raman scattering (SERS) technique by micro-Raman equipment has attracted great interest to combine information at the molecular level (Raman spectrograph) and morphology (optical microscopy), allowing to present such results also in the form of images. Thus, the combination of fluorescence microscopy (confocal in this case) and micro-Raman/SERS can be an important tool for detection of molecules of biological, pharmacological and/or environmental interest. In addition, SERS analyzes can also be used for the identification (in relation to interferers) and quantification of molecules. Thus, this research project has as the main objective to develop a methodology that allows applying the combination of confocal fluorescence microscopy and micro-Raman/SERS techniques in a complementary way to the same object of analyses (same sample). The development of the proposed methodology will have as the main object the analysis of giant unilamellar vesicles (GUVs), which is widely applied as a mimetic system of the lipidic structure of cell membrane. To these GUVs will be incorporated gold nanoparticles (AuNps, coated or not) containing dopamine (DA), an important neurotransmitter of the central nervous system. The AuNps are necessary to enhance the Raman signal (SERS) and the DA will act as the target molecule to be detected/ quantified inside and outside the GUVs. The combination of these microscopic/spectroscopic techniques is scarcely reported in the literature, being the principal innovator/challenging element of this project, which can be established as an important tool for studies of surface/molecule interaction, as well as the detection and quantification of interest molecules in biomimetic systems, or even be extended for studies in vitro and in vivo systems
