Abstract

Natural products of marine origin (PNM) emerge as a source of invaluable pharmacological potential. In this perspective, the sustainable supply of bioactive metabolites from the cultivation of marine microorganisms appears as an attractive solution for the rational prospection of PNM. Endemic Brazilian ascidia, Euherdmania sp., has been shown to be a promising source of microorganisms that produce cytotoxic substances. Previous studies have shown that the strain BRA-346, belonging to the genus Streptomyces, showed the most active extract with IC50 of 30ng/mL in human HCT-116 colorectal carcinoma cells. This extract also showed potent inhibition of the catalytic activity of the proteasome, by means of a target-directed assay using crystallography (IC50= 0.42ug/mL). Proteasome inhibitors are used clinically in the treatment of multiple myeloma, but have been tested with promising results in glioblastoma cells. Therefore, the objective of the present project will be to evaluate the mechanisms involved in cytotoxicity in glioblastoma cells and inhibition of proliferation of cells treated with the proteasome inhibitor (s) present in the BRA-346 extract.
