Abstract

Periodontal disease is an inflammatory disease that, when developed in a diabetic patient, manifests itself in an exacerbated manner, promoting the release of inflammatory mediators that promote bone resorption. The renin angiotensin system (RAS) is an endocrine axis that participates in various functions of the body, such as blood pressure control. The use of the type 1 angiotensin receptor antagonist in patients with periodontitis may have beneficial effects on disease progression, reducing inflammation and metalloproteinases, as well as on the formation and differentiation of osteoclasts. For this, 60 Balb/c mice will be used, divided into 6 groups: normal (N), diabetic (D), normal with periodontitis (N + DP), diabetic with periodontitis (D +DP), normal with periodontitis and treated with aliskiren (N + A + DP) diabetics with periodontitis treated with aliskiren (D + A + DP). The animals will undergo euthanasia 15 days after the ligation, and the gingival tissue surrounding the ligature will be removed for laboratory processing to study the inflammatory response. Thus, the objective of this work will be to investigate the role of renin on the production of Th17 cytokines, oxidative stress and antioxidants by the gingival tissue of diabetic mice submitted to experimental periodontitis. Immunohistochemical analyzes of IL-1beta, IL-6, TNF-alpha and IL-17 and the dosage of superoxide dismutase, catalase and glutathione peroxidase and malondialdehyde (MDA) produced by the gingival tissue of the animals will be performed. The results together will certainly contribute to the improvement of the knowledge of the cellular mechanisms involved in the inflammatory response, thus improving the treatment profile of the patients collaborating for the development of new therapeutics for the control of periodontitis in the presence of systemic diseases.
