Influence of tamoxifen on peri implant bone remodeling

The proposition of the present study is to avaliate the bone remodelation of osseointegrated implants in rat tibia, sistemic modified by Tamoxifen or not. 72 rats will be randomly distributed in 2 experimental groups (n=36), the animals will receive bilateral ovariectomy, implant installation in both tibias and after 8 weeks, Saline solution and Tamoxifen citrate will be administered by gavage for all the experimental period. Twelve animals of each group will be euthanized in the tenth, fourteenth e eighteenth week, both tibias will be collected and analyzed by histological, histomorphometric, immunohistochemistry and ultraestrutural analysis. The morphometric analysis consist in the description of the histologycal characteristics in the bone tissue around the implants threads and in the bone/implant interface. The immunohistochemical markings will be avaliated semi quantitatively for all the antibodies. For the histométric analysis the following criteria are to be adopted: The percentage of bone to implant contact and the percentage of the area of neoformed bone inside the limits of the implant threads, The analyzes will be performed on both sides of the implant, in the cervical, middle and apical thirds. The focus of the ultrastructural analysis will be the interface between bone and the surface of the implant. The relationship of osteocytes and their extensions with the surface of titanium and adjacent osteocytes, as well as the distribution of these cells in the peri-implant bone tissue will be evaluated. The quantitative data will be submitted to parametric or non-parametric statistical analyzes with significance level of 5%. (AU)
