Alveolar repair process evaluation in estrogen-deficient rats treated with OPG-Fc: immunohistochemistry, computerized microtomography and confocal microscopy analysis

Abstract

The present project aims to evaluate the alveolar repair process after the upper incisor extraction of ovariectomized rats that will be treated with OPG-Fc, mimicking the action of Denosumab, medicine used in anti-osteoporosis therapy. For the development of this project, 36 rats of 3 months will be divided into 3 experimental groups (SHAM, OVX/ST, OVX/OPG-Fc) that will be first submitted to ovariectomy or sham surgery. After 30 days, the rats will be submitted to treatment with OPG-Fc (10 mg/kg/2 times a week) maintained until the moment of euthanasia. The extraction of the upper right incisor will be performed 30 days after the start of treatment, being the period of analysis of the alveolar repair of 60 days after the exodontias. After euthanasia of the animals, according to the proposed experimental period, the jaws containing the alveoli under repair that will be submitted to computerized microtomography analysis; immunohistochemical analysis with labeling of the OPG, RANKL, TRAP and Osteocalcin proteins and epifluorescence microscopy. The quantitative data will be submitted to the normality curve for the definition of a parametric or non-parametric test, considering the level of significance of 5%. (AU)
