Abstract

Periodontal disease in a diabetic patient manifests itself in an exacerbated manner, promoting the release of inflammatory mediators that favor bone resorption by osteoclasts. Among the different cells involved in this process, mast cells are found abundantly in chronically inflamed periodontal tissues, favoring their participation in the pathogenesis of the disease. Few studies describe the role of miRNAs in the inflammatory response and bone metabolism during periodontal disease and fewer in the presence of systemic disease. Thus, the objective of the present study is to investigate the role of mast cells and their microRNAs on the inflammatory response and bone metabolism of diabetic mice submitted to experimental periodontitis. For this, Balb/c mice will be divided into 6 groups: normal (N), diabetic (D), normal with periodontitis (N + DP) diabetic periodontitis (D + DP), normal depleted mast cells with periodontitis (N + 48/80 + DP) and diabetic depleted mast cells with periodontitis (D + 48/80 + DP). Qualitative and quantitative (histomorphometric) analyzes of bone and inflamed tissue changes will be performed by means of light microscopy observations of H&E stained sections and immunohistochemistry for the targets related to the performance of mast cells, osteoblasts and osteoclasts. The combined results of the comparative analysis of the histomorphometric, immunological and molecular data will certainly contribute to the improvement of the knowledge of the molecular and cellular mechanisms involved in the inflammatory response necessary for the repair of the bone tissue, thus improving the treatment profile of the patients collaborating for the development of new therapeutics for the control of periodontitis in the presence of systemic diseases.
