Production of biohydrogen has the potential to be a renewable alternative to current technologies. Fermentative hydrogen production may be catalyzed by iron and nickel nanoparticles, as these chemical elements are cofactors of hydrogenase enzyme. This study aims to assess hydrogen production improvement when iron and nickel nanoparticles are added to the fermentative process. Central composite design will be applied to batch tests with initial pH (5, 6 and 7), initial substrate concentration (5, 15 and 30 g of glucose L-1) and nanoparticles concentration as entrance variables. Further, four Anaerobic Fluidized Bed Reactors (AFBR) (with iron nanoparticles, with nickel nanoparticles, with iron and nickel nanoparticles and without nanoparticles) will be operated on the optimal condition for hydrogen production obtained from batch tests. AFBR operational stages will consist in: (I) fed with glucose composite synthetic wastewater and (II) fed with real wastewater (vinasse). At the end of each stage the biofilm microbial community will be characterized by RNAr 16S sequencing. As an innovative research and for better process understanding, both reactors (batch and AFBR) will be inoculated with Clostridium butyricum pure culture.
