Treatment analysis of muscle injury by the association of suture and platelet rich plasma

Abstract

Muscle lesions are divided into mild, moderate and severe according to the amount of fibers ruptured. The standard treatment for severe injuries is surgery, in which the edges of the ruptured muscle are rejoined. Plasma Rich in Platelets (PRP) is another treatment that has positive results due to its growth factors that stimulate angiogenesis, chemotaxis and synthesis of new muscle fibers. In this way, the objective of this research will be to analyze the association of the two techniques in the muscular injury, hypothesizing that their union results in a more effective regeneration than the isolated application of the techniques. Seventy rats will be used in 5 groups: control (C), control lesion (CL), lesion and suture (LS), lesion and PRP (LP), lesion, suture and PRP. The injury process will occur through surgery in which the lateral portion of the gastrocnemius muscle will be sectioned. For the application of PRP, it will be extracted of the C group by means of cardiac puncture, and applied after 24 hours of injury. The animals will be euthanized on the 7th day after the injury. Seven animals will have the muscles submitted to the mechanical test while the others will have frozen muscle tissue for analysis of morphological characteristics, muscle collagen, fractal analysis, oxidative stress. For all analyzes will be performed normality test followed by appropriate post-tests depending on the presence of gaussian distribution or not. A significance level of 5% will be adopted. (AU)
