Abstract

Semen cryopreservation is a biotechnology with great potential for both poultry industry and endangered birds. However, this approach is still poorly explored because of the high susceptibility of the avian spermatozoa to the freezing process. Although reasons for such susceptibility have not been completely elucidated, previous studies assign plasma membrane damage as one of the possible factors leading to low freezing efficiency. Thus, the aim of this project is to evaluate the impact of fetal bovine serum (FBS) and cholesterol-loaded cyclodextrins on plasma membrane protection and spermatozoa fertility after the freezing-thawing process. To this end, semen samples from 15 Dekalb White males will be pooled and freezed with different FBS concentrations supplemented or not with 1mg/mL of cholesterol-loaded cyclodextrins. All semen pools will be assessed before and after cryopreservation for the following in vitro parameters motility, plasma and acrosome integrity, mitochondrial activity, lipid peroxidation and DNA fragmentation. Our long-term goal is to provide a freezing medium that confers some plasma membrane protection and, consequently, increases spermatozoa fertility after cryopreservation. (AU)
