THE ACTION OF MARESIN 1 (MAR1) ON FIBROGENESIS AND OSTEOGENIC ACTIVITY OF HUMAN PERIODONTAL LIGAMENT CELLS

Abstract

Maresins (macrophage mediators in resolving inflammation) are a family of specialized proresolving mediators of macrophage origin. These mediators are biosynthesized via initial lipoxygenation and molecular oxygen insertion at the carbon 14 position of docosahexaenoic acid (DHA) followed by epoxidation and oxygen incorporation at the carbon 7 position from a molecule of H2O. This research will evaluate how the maresin 1 (MaR1) can act on fibrogenesis and osteonenic activity of human periodontal ligament cells (hPDLCs) in an inflammatory microenvironment with cytokines IL-1² and TNF-±. Thus, we will evaluate the action of such mediators on the synthesis and gene expression of matrix metalloproteinases (MMPs) and their inhibitors (TIMPs), as well as its effects on collagen and osteogenic activity through alizarin red S staining and picro-sirius red staining. We will also perform analyzes about the expression of genes related to the periodontal ligament metabolism [(MMP-1, MMP-8, MMP-13, MMP-2, MMP-9, TIMP-1, TMIP-2, COL-1, ALP, osteopontin (OP), Periostin (PTN)], and the action of MaR1 on the phosphorylation of signal transduction molecules (MAPK and NF-ºB).
