Abstract

The boar semen is still little used and this occurs not only because the process causes reduction on the fertility rate. The current method of cryopreservation of boar semen is the Two Steps. This method is expensive and requires a long time, equipment and labor, fact that contributes to reduce the utilization of this biotechnology. This experiment aims to create simpler and faster methods that improves the post-thawed semen using smaller amounts of equipment and labor during the cryopreservation. Thus, two experiments will be realized. Experiment 1: Aims verify which Cryoprotectant penetrant agent (CPA) is better: 5 % Acetamide; 5% Dimethylformamide; 1% Glycerol and 4% Dimethylformamide; 2% Glycerol and 2% Dimethylformamide. The second experiment aims to compare the One Step method with the methodology commonly used in the boar semen cryopreservation, which is the Two Step. For this will be compared the best cryoprotecant found in the experiment 1 with the Two Step method Westendorf et al., 1975 and adapted by Yeste et al., 2016. After thawing, samples will be evaluated by motility characteristics (CASA), head's morfometry (ASMA), plasma and acrosomal membrane integrity, mitochondrial membrane potential, fluidity of plasma membrane lipids and lipoperoxidation by flow cytometer. Data will be submitted to analysis of mixed models (PROC MIXED), using the SAS (2002). (AU)
