Abstract

Synadenium grantii popularly known as "leiterinha" or "janaúba" is an euphorbiaceous shrub, native from West Africa, predominantly found in tropical and humid regions; rich in biologically active compounds such as flavonoids, alkaloids, lectins, esters, terpenes and others. In some regions of Brazil the populations use 18 drops of this latex in 1L of water for treatment of gastric diseases, allergies and cancer. An active principle of the latex (3,4,12,13-tetraacetylphorbol-20-phenylacetate) has already been identified as a rare diterpene ester phorbol responsible for the antiproliferative effect in tumor cell lines of gliomas, kidneys and lung cancer and triple-negative breast cancer, and has also been shown to be cytotoxic and antitumor to representative melanoma line B16F10. Our objective is to evaluate the antiproliferative properties of the latex of this plant in breast cancer cell lines, the second most common type of cancer among women in the world, MCF-7, is the representative of a less aggressive tumor type; and MDA-MB231 the representative of triple negative tumors, the most aggressive breast cancers in the clinics, with no specific therapy so far, evaluating growth curves, cell viability, migration and death, the formation of reactive oxygen species and the role of the hedgehog pathway in the inhibition of proliferation induced by Synadenium grantii, because this signaling pathway is responsible for cell proliferation and differentiation that has been associated with a worse prognosis for triple-negative breast cancer.
