Abstract

Semen preparation techniques in Assisted Reproduction laboratory are used to improve the sperm quality, fecundity and pregnancy rates. Often these techniques uses culture media supplementation with nutrients to enrich them and increase sperm survival. The present study aims to determine which culture media and / or supplementation contribute to sperm quality, from a sperm functional point of view, after incubation. To understand these relationships, a prospective study will be carried out using seminal samples obtained from 50 infertile patients referred to the Androscience Laboratory. Initially, routine seminal analysis and samples are classified in four groups, in according to culture media and supplementation: Group I - HTF comum media + SSS (control); Group II - Sperm Rinse Media + Fructose + Carnitine; Group III - G-IVF Plus Media + Fructose + SSS; Froup IV - CSCM Media + HSA. All samples are submitted to viability and hyperactive evaluation, radical oxygen species levels and integrity DNA test pre and post-incubation at 37°C CO2% environment.We will use Student's t test and analysis of variance (ANOVA) to compare study variables.
