Evaluation of bond interface in sound and caries-affected dentin under simulated pulpal pressure following aging in food-simulating solvents

Caries-affected dentin represents a field to be explored in order to its permanence after cavity preparation, according to the current conservative Restorative Dentistry philosophy. The aim of this study was to evaluate the bond strength and characteristics of adhesive interface in sound and caries-affected dentin, in the presence of simulated pulpal pressure and after aging in food-simulating solvents. Sixty sound human molars will be sectioned to obtain 2 mm thickness of dentin, from the highest pulp horn. The samples will be randomly divided into two groups (n=60), according to the type of substrate: sound dentin and caries-affected dentin. The preparation of the artificial caries-affected dentin will be performed by pH-cycling process. Under simulated pulpal pressure (15 cm H2O), all samples will be treated sequentially with 37% phosphoric acid, adhesive system (Universal Scotchbond, 3M/ESPE), light curing (10 s), build-up a composite resin block (Filtek Z350XT, 3M/ESPE) and light curing (20 s). Each group will be divided into 3 subgroups (n=20), according to aging food-simulating solvent: S1- distilled water, S2- 99% ethanol, S3- methyl ethyl ketone, at 37ºC for 24 h and 30 days. The samples will be submitted to microtensile bond strength test (10kgf load cell, 0.5m/min). Adhesive interface will be evaluated by scanning electron microscopy and transmission electron microscopy. Data will be submitted to an exploratory analysis to select the most adequate statistical test within the experimental conditions of this study (pd0.05). (AU)
