Antifungal effect of a fluconazole carrier nanosystem on Candida albicans and Candida glabrata

Abstract

The aim of this study is to synthesize a new fluconazole (FLZ) carrier nanosystem based on magnetic iron oxide nanoparticles (MNPs) and chitosan (CTS), as well as to evaluate the antifungal effect of this nanosystem against Candida albicans and Candida glabrata. The nanosystem will be prepared by loading of FLZ on MNPs coated with CTS and characterized by X-ray diffraction, infrared spectroscopy, thermogravimetric analysis and transmission electron microscopy. The minimum inhibitory concentration of the FLZ-CTS-MNP nanosystem capable of inhibiting the strains in planktonic state will be determined according to the microdilution method. After, biofilms of the two species will be formed (for 24 hours) into wells of 96-well plates and treated twice a day, during 2 days, with different concentrations of the nanosystem. Antibiofilm effect of the nanosystem will be evaluated by counting the number of cultivable cells, total biomass (crystal violet staining assay) and metabolic activity (XTT reduction assay). Appropriate controls will be included in all analyzes. Data will be analyzed to employ the most appropriate statistical test. P < .05 will be considered statistically significant. (AU)
