Interleukin-10 (IL-10) is an anti-inflammatory and immunosuppressive cytokine produced by several cell types, including macrophages, dendritic cells and various T-cell subgroups. This cytokine is released with the aim of modulating the synthesis of pro-inflammatory cytokines ensuring that no exacerbated inflammation occurs. However, polymorphisms in the IL-10 promoter region may be related to a "failure" in the control of inflammation, in addition to directly interfering with its level of expression, which may compromise the immune / inflammatory response. In addition to the above, infection with the gram-negative Helicobacter pylori bacteria in the gastric mucosa is associated with several gastric diseases, such as gastritis, ulcer, gastric carcinomas and seems to be involved in the synthesis of IL-10, especially Tregs and dendritic cells. Thus, the present project aims to characterize the polymorphisms and the gene expression of the Interleukin 10 gene in 250 samples of gastric biopsies of patients, using the PCR-RFLP technique and real-time PCR, respectively, in order to analyze and evaluate the frequency of promoter-1082 (G/A) and -3575 (A/T) polymorphisms as well as their haplotypes. Since IL-10 polymorphisms may interfere with the immune response against H. pylori bacteria, the development of this design could lead to new or better understandings of the molecular mechanisms of gastric diseases (AU)
