Bothrops envenomation often includes local effects and systemic reactions represented mainly by coagulation disturbances. SVSPs, SVMPs, PLA2s and CTLs are main protein families involved in these disturbances. These toxins are able to act on components of the coagulation cascade, promote hemorrhage and interfere by different ways in the formation of the hemostatic plug and platelet aggregation. Recently we characterized proteomes and biological activities of four B. atrox venom pools obtained from snakes collected at different habitats at west of Pará State, Brazilian Amazon. We showed that Bothrops antivenom neutralized hemorrhage induced by all the venoms, but did not completely neutralized coagulant activity of the várzea venom. Then, due to marked variability of the venom, considering also the importance of the hemostatic disorders in the envenomation by B. atrox, purified toxins able to act on coagulation system, from venom pools of snakes from different locations. Fractions composed by SVSP and PLA2s were obtained with reasonable homogeneity and preserved biological activity. However, in our lab we do not have an array for comprehensive analysis of hemostatic disturbances, on the other hand, Dr. Bryan Fry's lab (University of Queensland, Australia) have a sophisticated platform that will enable us to perform a more comprehensive characterization of the hemostatic disturbances induced by B. atrox venoms and some of their components (SVSPs and PLA2s). The results obtained in these analyzes may provide new insights for improvement of the serum therapy of envenomation at Brazilian Amazon. At same time, the internship at Dr. Fry's lab can open new opportunities for collaborative studies between our group (Instituto Butantan) and researchers from University of Queensland, Australia.
