Kidney expression of markers of injury and apoptosis in rats subjected to renal ischemia and reperfusion and treatment with Brazil nuts

Abstract

The ischemia and reperfusion (IR) is a condition that occurs during the procedures involved in the transplantation of organs, which consists in one of the causes of acute renal failure of the graft. Despite advances in the area, mortality due to acute renal failure is high. Study developed in our laboratory by students (FAPESP processes 2015/08232-3, 2015/18313-0 and 2015/25616-0) showed that pretreatment with 75 mg of Brazil nuts improves renal function parameters changed by IR, with elevation of creatinine clearance and urinary flow, reduction of proteinuria and the plasmatic urea, as well as reducing the renal expression of iNOS, nitrotirosine and the influx of macrophages. However, we did not find significant difference in renal histopathological analysis between the IR and control groups. Objective: to analyse the action of Brazil nuts on apoptosis and kidney injury of rats submitted to IR process. Methods: the fragments of kidney tissues of male Wistar rats were collected previously, included in paraffin and stored in our laboratory. In these experiments, the animals were divided into six groups (06 rats/group), euthanized after 48 hours of surgical procedures, as follows: SHAM (surgery without arterial clamping); SHAM + C75 and SHAM + C150 (75 or 150 mg of Brazil nuts for 7 days before surgery SHAM); IR (IR surgery with renal artery clamping by 30 min.); IR + C75 and IR + C150 (75 or 150 mg of Brazil nuts for 7 days before IR surgery). In the present project, the renal fragments will be sectioned and submitted to immunohistochemical analysis of expression of caspase-3 for evaluation of apoptosis, and expression of vimentin and alpha-actin for assessment of renal injury.
