Abstract

Neotropical deer have been enduring population decrease mainly due anthropic actions. In this context, tools such as genomic banks are important strategies for maintenance of genetic diversity. Testicular tissue could be an interesting source for composing these banks and preserving material for subsequent propagation of individuals. To achieve this, it is necessary to develop cryopreservation protocols that fits to tissue specificities using cryoprotectants in the appropriate concentration, allowing the cells to remain viable after thawing. The low number of individuals of wild species makes necessary to use experimental models such as bovine, once that belong to a taxonomic group that is close and suitable for deer. Therefore, the objective of this work is to evaluate the cryopreservation of testicular tissue of bovine by the slow-freezing method. Seven animals will be used and three cryopreservation media will be tested consisting of DMEM and 20% BFS in addition to 10% DMSO or 15% DMSO or 10% glycerol. Cell viability will be accessed in fresh and post-thaw samples using trypan blue, and the cells will be counted in Neubauer chamber. (AU)
