Antimicrobial activity of different protocols of irrigation in grooves produced in bovine dentin: study on laser scanning confocal microscopy

Abstract

The aim of the present study will be to evaluate in vitro in bovine dentin antibiofilme activity promoted by different irrigation systems. The groups to be tested will be the following (n = 10): Group 1-RinsEndo System; Group 2-Ultrasound (irrissonic); Group 3 - Easy Clean alternate; Group 4 - Easy Clean Rotary; Group 5 - conventional (syringe and cannula). The irrigating solution used is the 6% sodium hypochlorite. 50 sterile dentin blocks 2 x 2 mm shall be used. To induce the Dentinal infection, the dentin blocks shall be laid down in a plate of orthodontic Hawley. The block of dentin in contact with the oral cavity shall be fixed at 1 mm below the surface of the Board to enable the accumulation of biofilm. The card is being used by a volunteer for 72 hours for induction of dentin infection. After the infection process, each block will be incubated in culture medium BHI, 37oC, for 48 hours under conditions of aerobic organism. The blocks infected will be adapted to perforations of 2.5 x 2 mm 3 mm from the apex of the incisors. The infected side is placed toward the root canal. The diameter of the root canal will be standardized using Gates Glidden BUR # 5 (1 mm) in the apical level and the length of the channel will be standardized at 12 mm. Will be applied a final Protocol of irrigation during 1 minute. The following techniques will be used. The blocks will be dehydrated with alcohol, metalized with Platinum and mounted on foundations for later analysis in the scanning electron microscope 15 randomly selected areas will be scanned each block, using an increase of 2,400 X. In total, 150 areas will be assessed for each group. For quantification shall be used modified scores following the recommendations of Bhuva et al. (2010). For the study of the comparison between the time and the experimental groups will be applied to analysis of variance (ANOVA) followed by the Tukey test for multiple comparisons between them. For analysis of concordance between the evaluators will use the Kappa test. The level of significance will be 5%. (AU)
