Abstract

Anthropic actions on the environment, as the introduction of metals in water can cause genotoxic and cytotoxic effects on animals, and affect physiological processes such as reproduction. The toxicity of these metals, including aluminum (Al) can be potentiated when in synergy with high temperatures, influencing fish health and the quality of gametes, threatening the success of the reproduction. The hypothesis of this proposal is that Al, in synergy with the high temperature, can affect the seminal plasma and hence freezability of spermatozoa. The effects of exposure to Al will be assessed at different water temperature, on cytogenetic patterns, sperm characteristics, protein profile of seminal plasma and semen freezeability of Astyanax altiparanae. Sexually mature male A. altiparanae will be subdivided into nine groups: six groups without addition of Al, in acidic or neutral water pH in three different temperatures, 20, 25 and 30°C respectively, and the other three groups containing Al in the water (0.5 mg.L-1) at acidic pH, in the same temperatures described. The study will be carried out in two stages: 1) the animals exposed to the above conditions for 24 hours; and 2) the animals exposed for 96 hours; after each of these times, all animals from the tank will be sampled (n = 8) to run the toxicity tests, semen analysis and protein profile of seminal plasma. The blood will be collected to perform the micronucleus test and the comet assay. For seminal analysis, the animals will be induced to spermiation and subsequently the semen will be collected to analyze the osmolarity, pH, volume, morphology, kinetics, concentration, comet assay, micronucleus test, sperm ultrastructure and protein profile of the seminal plasma. The semen will be cryopreserved and semen quality parameters will be re-evaluated after thawing. Al content will be measured in gonads and semen. (AU)
