In necropulpectomies, the main function of intracanal medications is to combat microorganisms that were not eliminated during the biomechanical preparation of the root canals. Calcium hydroxide medication has been widely used, but is not effective in eliminating Enterococcus faecalis, mainly in the form of biofilm. This study aims to evaluate whether the use of N-Acetylcysteine or Ambroxol Hydrochloride prior to medication, would potentiate its antimicrobial action. Material and Methods: The experiments will be divided into 6 groups according to the medications in question: G1 - Ambroxol Hydrochloride (5 minutes); G2 -N-Acetylcysteine (5 minutes); G3- Ambroxol Hydrochloride (5 minutes) / Calen PMCC; G4 - Ambroxol Hydrochloride (2 five minute applications) Calen PMCC; G5-N-Acetylcysteine (5 minutes) / Calen PMCC; G6-N-Acetylcysteine (2 five minute applications) / Calen PMCC; G7- Control. Antimicrobial action: Over 70 blocks of bovine dentin will be induced in vitro biofilm growth of Enterococcus faecalis. After the development of this, the contact test will be carried out according to the group. For microscopic analysis, each block will be stained by Live / Dead staining and the percentage of live cells measured by confocal scanning laser microscope and bioimage program. (AU)
