Biological performance of spongin extracted from marine sponges composite enriched with biosilicate and combined or not with laser therapy on bone tissue engineering

Abstract

Bone fractures are a major challenge for clinical practice and it is related to high rates of morbidity and mortality. In this context, the bone tissue engineering has been developing new biomaterials and therapeutic resources in order to optimize bone repair. Therefore, the objectives of this study are: (i) to produce and characterize new composites based on spongin extracted from marine sponges enriched with Biosilicate®, associating or not with low level laser therapy (LLLT); (ii) to evaluate the biological responses, in vitro, induced by sponging and Biosilicato®/spongin composite and LLLT, and (iii) to assess, through in vivo tests, the composite effects, combined or not with LLLT, using a calvarial bone defect model in rats. For this purpose, the composites will characterize by scanning electron microscope (SEM), X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR). For the in vitro and in vivo test will be used these experimental groups: Control; Biosilicato®; Spongin; Biosilicato®/spongin; Control + LLLT; Biosilicato® + LLLT; Spongin + LLLT; Biosilicato®/spongin + LLLT. For the in vitro tests, the cytotoxicity of the composites, combined or not with LLLT (808 nm; 30 mW; 1.07 W/cm2; 10 J/cm2 and 9 seconds), will be evaluated by Alamar Blue assay using MC3T3 and L929 cells after 24, 72 and 120h. Furthermore, alkaline phosphatase tests, MTT, and analysis of mineralized nodules will be performed. For the in vivo tests, 128 Wistar rats will be submitted to a surgical procedure in order to induce a calvarial bone defect model. The defect will be filled with the biomaterial, associated or not with LLLT (808 nm; 30 mW; 1.07 W/cm2; 30 J/cm2 and 28 seconds). Thus, each experimental group will be divided into 2 subgroups, euthanized after 15 and 45 days post-surgery. It will be performed of histopathology, morphometry, micro-CT and immunohistochemistry analysis. This study will contribute with the development of new technologies, obtained from marine resources, for improving bone repair treatments, using national and natural biomaterials associated with LLLT.
