Evaluation of dentifrices with açaí berry and/or fluoride on the prevention of erosion associated to dentin abrasion

Abstract

The objective of this study is to investigate the protective effect of experimental toothpastes containing extract of acai-berry and fluoride on dentin erosion associated to abrasion in preservation of demineralized collagen matrix degradation. The in vitro study will conducted using bovine dentin blocks (n = 12 / group) that will be exposed to pH cycles, associating abrasion by brushing in automatic machine. The groups will be according compound with the active agent present in toothpaste: toothpaste containing acai-berry extract (10%), fluoridated toothpaste (1100 ppmF, as NaF) or both one combination. More two groups will be added represented for placebo dentifrice (negative control) and chlorhexidine dentifrice (positive control). The dentin blocks will be submitted to four de-remineralization per day, repeated for 5 days. The demineralization will be caused by 1% citric acid (90, 4x / day) and an artificial saliva to remineralization (1h, 4x / day). Among repetitions of pH cycling, the dentin blocks will brushed, using automatic brushing machine and dentifrice solution (3 g dentifrice / 10 mL water) for 15 s after one first and the last demineralization. Dentin loss will be assessed by contact profilometry. In sequence demineralised organic matrix (DOM) will be removed by immersing the dentin blocks in solution containing collagenase obtained from Clostridium histolyticum type VII (100 U / ml) for 96 hours at 37 ° C and profilometric analysis will be evaluated in the absence of collagen fibrils. The thickness of the DOM will be calculated the difference between the values obtained in the second and first profilometry. Data will be analyzed by ANOVA (p <0.05). (AU)
