Evaluation of thyroid hormone action upon noncoding RNA expression in osteoblastic cells derived of adipose tissue

Abstract

The bone tissue is extremely complex and is regulated by systemic and local factors. It presents considerable metabolic activity including the removal of mineralized bone through the osteoclasts followed by the formation of bone matrix by the osteoblasts. The T3 might lead to a complex response to the bone tissue activity and produces effects upon bone resorption as well as bone formation. Therefore, various studies aim at verifying the hormone action upon bone metabolism and within them there are groups isolating mesenchymal stem cell from human adipose tissue and differentiating into osteoblasts. The differentiation might be triggered by several factors including the expression of some specific genes that are related to various gene expression regulating mechanisms such as long non-codingRNAs (lncRNAs). Hence, the identification of long RNAs involved in the osteoblast differentiation from the quantitative transcriptome sequencing might lead to answer some questions about the complex bone metabolism. Thus, the present study allows the analysis both the differentiation of osteoblasts and the profile of transcriptome during the progression to osteoclastogenesis stimulated by thyroid hormone. It also identifies long RNAs with potential applicability to the reversion or maintenance of such processes in a field that is eminent within bone biology. (AU)
