Abstract

Recent studies have shown that the water deprivation (WD) induces an increase in the abundance of the gonadotropin inducible ovarian transcription factor 1 (Giot1) mRNA expression in the supraoptic nucleus (SON) of hypothalamus involved on hydromineral homeostasis. It's wide knowledge that, for example water deprivation, activates hypothalamic neurons inducing the production and release of vasopressin (AVP) and oxytocin (OT), as well as increasing drink ingestion. Interestingly, AVP and OT release in response to WD was further increased after oestradiol (E2) replacement in female ovariectomized rats. Moreover, oestradiol also reduce water intake in WD, showing that E2 participates in hydromineral regulation. In addition, data from literature shows that, centrally E2 acts in specific hypothalamic pathways, but the E2 mechanisms involved on AVP and OT secretion are still unknown. In these context, this project proposes evaluate the relationship between oestradiol and Giot1 in CNS. To investigate the role of Giot1 on the hormonal release and water/saline intake, we will use ovariectomized rats with E2 replacement, summited to water deprivation. Furthermore, a lentiviral vector will be used to knockdown Giot1 in SON and PVN nuclei. In this set of animals, hormonal and behavioral parameters will be evaluate, as well as the central RNAm expression of various neuromodulators involved on hydromineral homeostasis.
