Abstract

The tissue engineering combined with molecular biology are associated in the treatment of numerous diseases, aiming to restore healthy by regeneration or reparation. For this, scaffolds can be used in Dentistry, particularly in Endodontics, as the basis for tissue healing. In this study will be performed a 3D tapered PCL scaffold, with or without reinforcement with calcium silicate, addition or without antimicrobials (metronidazole and ciprofloxacin). It will be performed the preparation and characterization of scaffolds by eletrospinning, SEM assay and liquid chromatography. Among the biological analysis, the scaffolds will be evaluated regarding cell adhesion and proliferation tests will be carried out atomic force microscopy associated with 3D fluorescence confocal microscopy, cytotoxicity assays (MTT test) genotoxicity (micronuclei test) on dental pulp stem cells (DPSC). It will also be evaluated the expression of alkaline phosphatase activity (ALP), DMP-1 (matrix protein 1), DMP-2 (matrix protein 2), and DSPP (dentin sialophosphoprotein) using a 3D fluorescence confocal microscopy associated with microscopy atomic force. The analysis of antimicrobial activity against Enterococcus feacalis will be evaluated. All tests will be performed in triplicate. Statistical analyzes will be performed by ANOVA, Tukey and Mann-Whitney test (significance level of 5%).
