The chemistry of natural products has remained as a source of inspiration for the discovery of new drugs. Due the enormous diversity of secondary metabolites and biological activities that can be found in the nature, many drugs in the current pharmacopeias are of natural product origin or derivatives of them. However, classical bioassay-guided fractionation using preparative-scale isolation followed by testing using traditional bioassays are time-consuming and complicated when the final yield of pure compounds is low. Consequently, new effective technologies for screening plant extracts have been developed to yield more rapid results against several therapeutic targets. High-resolution bioassays coupled with HPLC-HRMS-SPE-NMR has been an important new development for identification of bioactive constituents directly from crude plant extracts. Therefore, the main goal of the proposed internship project is to evaluate the high-resolution a-glucosidase, a-amylase and monoamine oxidases inhibition profiles as well as to obtain structural characterization of all constituents from Picramnia. In our previous studies with Picramnia glazioviana, we demonstrated the isolation of new nor-triterpenes, but some NMR experiments as well as the insecticide and fungicide bioassays with these pure compounds was not obtained mainly due to the low amount isolated of them. Consequently, evaluating the extract rich in these new compounds by high-resolution bioassay/HPLC-HRMS-SPE-NMR brings expectation to complete their structural elucidation as well as to find out other important bioactivity for these compounds. We expect also, this abroad internship under the supervision of Prof. Dr. Dan Staerk will immensely contribute and enlarge our knowledge about this relevant technology. (AU)
