Abstract

In a previous study, our research group demonstrated that 4% TiF4 varnish did not show antimicrobial effect in a microcosm biofilm on enamel; however, it was able to decrease enamel demineralization. Therefore, the present study aims to evaluate the effect of this agent on the microbial viability, acid and extracellular polysaccharide (PEC) production as well as on the microorganism counting of a cariogenic microcosm biofilm produced on root dentine. For the microcosm biofilm formation, saliva will be collected from 10 healthy subjects, who must have not brush their teeth for the last 24h and not drink or eat for the last 2h. The saliva will be diluted (70% saliva and 30% glycerol), and aliquots of 1ml will be stored at -80ºC. Sample of bovine dentine (4mm x 4mm) will be prepared and treated for 6h with the varnishes containing: A) 4% TiF4 (pH 1.0, 2.45% F); B) 5.42% NaF (pH 5.0, 2.45% F); C) placebo D) 2% Chlorexidine gel - positive control or E) untreated - negative control (n=4 x biological triplicate/assay). These samples will be plated in 24-wells plates and exposed to the human saliva mixed with McBain saliva (1:50) with 0.2% sucrose for the microcosm biofilm formation. In the Assay 1, the live and dead bacteria will be evidenced by fluorescence using a confocal microscope. In the Assay 2, acidogenicity will be evaluated, by measuring the lactic acid using enzymatic methods and microplate reader. In the Assay 3, the quantity of PEC production will be evaluated by colorimetric method using microplate reader. In the Assay 4, the microbial suspension removed from the biofilm will be diluted and spread in Agar plates for colony forming unit (CFU) counting for total microorganism, total streptococcus, mutans streptococcus and total lactobacillus and in the Assay 5, the dentine demineralization will be measured using transverse microradiography. The data will be plotted and submitted to the statistical analyses (p<0.05). (AU)
