The aim of this study is to synthesize a compound containing silver nanoparticles (Ag-NP), sodium trimetaphosphate (TMP) and fluoride (F), characterize it and evaluate the antimicrobial action in planktonic cells of Candida albicans and Streptococcus mutans. For this, Ag-NP will be synthesized by reduction of silver nitrate using a chemical route in the presence of F and TMP. This compound will be characterized using scanning electron microscopy (SEM) and diffraction X-ray (XRD). It will also be assessed the amount of silver ions through ion-specific electrode. The antimicrobial action in planktonic cells will be evaluated by the minimum inhibitory concentration (MIC) for Streptococcus mutans ATCC 25175 and Candida albicans ATCC 10231 using the microdilution method. Each microorganism suspension (100 uL) will be added to the microtiter plates wells (96 wells) containing 100 uL of each concentration of specific compounds. Subsequently, the microtiter plates will be incubated at 37 ° C and MICs are determined visually as the lowest concentration able to inhibit the growth of microorganisms after 48 hours of incubation for Candida albicans and 24 hours for Streptococccus mutans. All tests will be performed in triplicate on three different occasions. (AU)
